蛋白印迹 (Western Blot) 是根据抗原抗体的特异性结合检测复杂样品中的某种蛋白的方法，用于检测经电泳或斑点杂交后固定于膜上的蛋白质。由于Western blot检测技术结合了凝胶电泳的高分辨率和固相免疫测定的特异敏感等多种特点，可检测到1-5 ng中等大小的靶蛋白，成为蛋白分析的一种常规技术，对蛋白进行定性和半定量分析。

实验样品要求：
（1）细胞不少于5×106个；
（2）组织样品不少于50mg；
（3）总蛋白浓度不低于 2μg/μl（至少50μl）；
注意事项：
（1）每张膜上做一种目的蛋白和相应的内参蛋白。每张膜上可以做8个样品。一般状况下，2周即可完成实验。如遇特殊情况，如一抗不好，蛋白浓度低或蛋白特殊，如分子量大等，周期会有所延长。
（2）Western
Blot细胞蛋白样本的准备及运输：去掉培养液，用PBS洗一次后，加入少量PBS，用细胞刮将细胞刮下，用1ml枪吸取细胞易于1.5ml EP管中，快速离心（升至最高转速就可stop）去除上清后，可按下面两种方法运输：A.细胞干运输：将细胞干放入液氮速冻，-80℃保存; 如需长途过夜运输，建议干冰运输；如当天送达样本，可用液氮或者-80冰袋运输；B.细胞裂解液运输：加入一定量的胞裂解液后即可冷藏运输（冰袋或者冰盒）。如需检测磷酸化蛋白，建议裂解液中加入终浓度1mM的NaF，NaF可抑制磷酸酶活性，保护蛋白磷酸化水平。如果加入裂解液后提取蛋白，则置于冰上裂解1 h，12000rpm离心10 min后取上清，即为蛋白样本。蛋白样本可-20度冷藏运输（建议快递寄送用干冰或者足量-80度冰袋），保存需要-80℃。
（3）蛋白样本运输：蛋白样本需要经过浓度测定，再进行变性，变性时先加入Sample buffer，再95℃ 10 min，Sample buffer可防止变性导致的蛋白沉淀析出。变性的蛋白可直接冷藏运输，-20℃稳定保存。
