1,选择A和B中共有的酶切位点a和b（如果A中找不到合适的酶切位点可用通过PCR的方法添加）2,分别用酶a和b酶切A和B片段.3,回收酶切后的A何B片段,用T4连接酶室温连接过夜或者快速连接酶16℃连接30min.4,转化,将A和B的连接混合物转化到大肠杆菌感受态细胞中,在相应抗性的培养基中培养12-18h至培养板中长出单菌落.5,挑取菌落空斑至液体培养基中培养12-16h；（此步骤之前也可用蓝白斑删选的办法删选培养板中的单菌落）.6,小量抽提连接产物质粒,用酶a和b酶切鉴定,如果酶切产物为两条大小和A、B片段大小吻合的条带,则连接成功,此步骤如果为进一步确定结果,还可再选用一组其他酶做酶切鉴定.
对在2支培养管中培养的相同（品系、数量）细胞分别给予相同时长的不同刺激，现欲知某基因A在两管细胞中的转录表达是否存在差异。请作实验设计达成此目的。
